ELDISINE est généralement déconseillé en association avec les vaccins vivants atténués, la phénytoïne, la fosphénytoïne et avec l'itraconazole.
ELDISINE ne doit en aucun cas être associé à une vaccination contre la fièvre jaune et à la phénytoïne.
Une contraception efficace doit être mise en place après la vérification de l'absence de grossesse chez la femme.
ELDISINE ne doit pas être injecté par voie intrathécale, sous arachnoïdienne, ou intrarachidienne.
Des contrôles de l'hémogramme doivent être effectués régulièrement, des précautions doivent être prise en cas de cardiopathie ischémique.

Une surveillance accrue doit être mise en place en cas d'association avec les anticoagulants oraux.
