Le principe actif de DOXORUBICINE inhibe la synthèse de l'ADN et de l'ARN en s'intercalant dans sa structure donc la multiplication cellulaire. Il fait partie de la famille des anthracyclines.

Effets secondaires
On observe fréquemment une fatigue, et un état fébrile généralisé qui disparaît par la suite. La doxorubicine provoque également des nausées, des vomissements, des douleurs abdominales et des diarrhées.
En raison de la cardiotoxicité de DOXORUBICINE, des douleurs disséminées et une oppression thoracique, des bouffées de chaleur, des troubles du rythme peuvent survenir durant le traitement. Une immunodépression est également observée, elle est due à la destruction des cellules des défenses immunitaires dans le sang et est réversible après le traitement. De plus, il arrive également que le traitement déclenche des leucémies secondaires.
DOXORUBICINE provoque une perte des cheveux dans 90% des cas (alopécie) elle est réversible à l’arrêt du traitement.

Contre-indications / Précautions d'emploi
DOXORUBICINE est contre-indiqué en cas d'allergie à l'un de ses composants, chez les personnes atteintes d'une cardiopathie avec insuffisance myocardique, en cas de grossesse, d'allaitement.
Dans tous les cas, une surveillance cardiaque et hématologique est nécessaire durant le traitement. Une numération, une formule sanguine et un électrocardiagramme seront réalisés avant chaque injection.
La surveillance doit être accrue en cas d'insuffisance hépatique, et de radiothérapie.
Interactions médicamenteuses
DOXORUBICINE ne doit pas être administré avec le cyclophosphamide car cela augmente les effets cardiotoxiques de la doxorubicine et le risque de cystite hémorragique du au cyclophosphamide. Il ne doit pas non plus être administré avec la mercaptopurine car cela augmente la toxicité hépatique (foie) provoquée par la mercaptopurine.
Il est également préférable de ne pas associer la doxorubicine à la ciclosporine et aux vaccins vivants atténués. Préférez un vaccin inactivé s'il existe.
